Research Overview

Tesamorelin is a synthetic analog of growth hormone-releasing hormone (GHRH) incorporating the full 44-amino acid sequence of human GHRH with a trans-3-hexenoic acid modification at the N-terminus to improve stability. It is the most clinically studied GHRH analog and provides a well-characterized tool for GH axis research.

Mechanism of Action

Tesamorelin binds to GHRH receptors on somatotroph cells of the anterior pituitary, stimulating pulsatile release of endogenous growth hormone. GH then stimulates IGF-1 production in the liver, driving anabolic and lipolytic effects. The trans-3-hexenoic acid modification protects the N-terminus from dipeptidyl peptidase IV (DPP-IV) cleavage, extending activity compared to native GHRH.

Experimental Applications

Research includes pituitary GH axis studies, IGF-1 modulation, visceral fat reduction models, lipodystrophy research, GH deficiency models, cognitive function and neuroprotection, cardiovascular risk marker analysis, and GH pulsatility studies.
