MOTS-c is a 16-amino acid peptide encoded in the mitochondrial 12S rRNA gene, representing a new class of mitochondrial-derived peptides (MDPs). It is a potent metabolic regulator that translocates to the nucleus under stress conditions and modulates gene expression related to metabolic homeostasis and aging.
MOTS-c activates AMPK (AMP-activated protein kinase) signaling to enhance glucose uptake, fatty acid oxidation, and mitochondrial biogenesis. Under metabolic stress it translocates to the nucleus where it functions as a transcription regulator, modulating the folate cycle and reducing reactive oxygen species. It also influences the one-carbon metabolic pathway and shows anti-inflammatory and anti-aging properties in aging models.
