IGF-1 LR3

IGF-1 LR3 (Long Arg3 IGF-1) is a synthetic analog of insulin-like growth factor-1 with an arginine at position 3 and a 13-amino acid N-terminal extension that dramatically reduces IGFBP binding affinity. This results in a ~3× longer half-life than native IGF-1 and enhanced potency, making it a powerful tool for IGF-1 receptor biology research.

IGF-1 LR3 binds IGF-1R (IC50 similar to native IGF-1) and activates the PI3K/Akt/mTOR and MAPK/Erk signaling cascades, driving protein synthesis, cell proliferation, and survival. The Arg3 substitution and N-terminal extension prevent binding to insulin-like growth factor binding proteins (IGFBPs), meaning the compound circulates in its free, biologically active form for longer, producing sustained anabolic signaling.
