Melanotan II is a cyclic heptapeptide analog of alpha-melanocyte-stimulating hormone (α-MSH) with enhanced receptor binding and metabolic stability. It is a non-selective agonist at MC1R through MC4R and is used extensively in melanocortin receptor pharmacology and skin pigmentation research.

Mechanism of Action

Melanotan II activates all five melanocortin receptor subtypes. MC1R stimulation in melanocytes increases eumelanin production (skin/hair darkening). MC3R and MC4R activation in the hypothalamus suppresses appetite and modulates sexual arousal. MC2R stimulation drives adrenal cortisol output. The cyclic structure confers higher receptor affinity and metabolic stability compared to linear α-MSH fragments.
