Melanotan 1 (Afamelanotide) is a linear 13-amino acid synthetic analog of α-melanocyte-stimulating hormone with a norleucine at position 4 and D-phenylalanine at position 7 (NDP-α-MSH), conferring greater potency and stability than native α-MSH. It is one of the most MC1R-selective melanocortin analogs and is studied for photoprotection and pigmentation research.
Melanotan 1 binds preferentially to MC1R on melanocytes, activating adenylate cyclase and increasing cAMP, which drives MITF transcription factor activity and upregulates enzymes in the eumelanin biosynthesis pathway (tyrosinase, TRP-1, TRP-2). This produces UV-independent melanogenesis. At higher concentrations it also activates MC3R and MC4R, producing appetite and photoprotective effects.
Research includes melanogenesis and pigmentation biology, photoprotection mechanisms, MC1R-selective pharmacology, DNA damage repair stimulation in skin, erythropoietic protoporphyria models, comparative studies against cyclic melanocortin analogs, and skin cancer photoprotection research.
